RESUMEN: El tizón de la hoja es una enfermedad que afecta al cultivo de la cebolla y una alternativa biológica para su manejo es el uso de hongos antagónicos del género Trichoderma. Debido a que los estudios sobre la identificación del agente causal del tizón foliar en la cebolla cultivada en México son escasos, fue el objetivo de esta investigación. Asimismo, se evaluó la actividad antagónica de Trichoderma asperellum TC3 para el control del agente causal de la enfermedad. Se recolectaron cien hojas de plantas de cebolla con síntomas de tizón de la hoja en “Chiautla de Tapia”, estado de Puebla, México. Los fragmentos de tejido se cultivaron en medio de papa, dextrosa y agar (PDA) y las colonias de hongos se clasificaron en tres grupos según sus características morfológicas. Las colonias de hongos de los grupos 1 y 2 no presentaron esporas. Sin embargo, un aislado (DC) obtenido del grupo 2 se identificó
molecularmente, mientras que tres cultivos monospóricos (SV1, SV2 y SV3) se obtuvieron del grupo 3 y se realizó su identificación morfológica y molecular, y se usaron en las pruebas de patogenicidad. La identificación morfológica de los aislados se realizó mediante claves taxonómicas. La identificación molecular del aislado DC y los tres aislados SV se llevó a cabo mediante la amplificación de las secuencias de regiones del transcriptor interno espaciador ITS con los oligonucleótidos ITS1 y ITS4. Además, para SV1, SV2 y SV3 se amplificaron las secuencias de las regiones del gen gpd de la enzima gliceraldehído 3-fosfato deshidrogenasa utilizando oligonucleótidos diseñados en este estudio. La actividad antagónica in vitro de T. asperellum aislada de cultivos de mango (Tm), tomate (Tt) y cebolla (TC3) contra el patógeno se evaluó mediante ensayos de cultivo dual y medio enriquecido con filtrados de cultivo de cada aislado de T. asperellum. Las características morfológicas y la identificación molecular indicaron que los aislados de SV1, SV2 y SV3 corresponden a Stemphylium vesicarium; mientras que el análisis molecular de las secuencias ITS del aislados de DC mostró similitud con hongos del género Alternaria. La infección con los aislados SV1, SV2 y SV3 de las hojas provocó la formación de manchas blancas, clorosis, lesiones de color negro, colapso, necrosis y muerte del tejido foliar. El ensayo de actividad antagónica con medio enriquecido con filtrados de cultivo de cada uno de los aislados de T.asperellum indicó que el aislado TC3 de T. asperellum inhibió el crecimiento micelial de S.
vesicarium en 66.9%; seguido de los aislados Tt y Tm con 43.5 y 37.2%, respectivamente. Los aislados TC3 y Tm de T. asperellum también inhibieron la producción de conidios de S. vesicarium en un 40 y 45%, respectivamente. Los tres aislados de T. asperellum inhibieron el crecimiento micelial de S. vesicarium en cultivo dual; el aislado de Tm mostró el porcentaje más alto de inhibición del crecimiento micelial (61,1%), seguido de los aislados de Tt y TC3 de T. asperellum con 56,4 y 54,4%, respectivamente. En conclusión, en la cebolla cultivada en el estado de Puebla, México, uno de los agentes causales del tizón foliar es S. vesicarium y podría estar asociado con especies de Alternaria. El aislado de TC3 de T. asperellum obtenido de cultivos de cebolla podría ser una alternativa para el control de S. vesicarium. Este es el primer estudio que reporta la presencia de S. vesicarium en cebolla cultivada en México y que para la identificación de la especie combina datos morfológicos y moleculares.
ABSTRACT: The leaf blight is a disease that affects the onion crop and a biological alternative for itsmanagement is the use of antagonistic fungi of the genus Trichoderma. Due the studies on the identification of the causal agent of leaf blight in onion cultivated on Mexico are scarce, it was the aim of this investigation. Likewise, the antagonic activity of Trichoderma asperellum TC3 for the control of the causal agent of the disease was evaluate. One hundred leaves of onion plants with symptoms of leaf blight were collected in “Chiautla de Tapia”, state of Puebla, Mexico. The tissue fragments were cultivated in potato, dextrose and agar (PDA) medium and the fungal colonies were classified in three groups according to their morphological characteristics. The fungi colonies of groups 1 and 2 did not present spores. However, one isolate (DC) obtained from group 2 was identified molecularly, whereas three monosporic cultures (SV1, SV2 and SV3) were obtained from group 3 and was realized their morphological and molecular identification, and used in the pathogenicity tests. The morphological identification of the isolates was made using taxonomic keys. The molecular
identification of the isolate DC and the three SV isolates was carried out by the amplification of the regions sequences of the internal transcriber spacer ITS with the primers ITS1 and ITS4. Additionally, for the SV1, SV2 and SV3 isolates were amplified the sequences regions of gpd gene of the glyceraldehyde 3-phosphate dehydrogenase enzyme utilizing primers designed in this study. The in vitro antagonic activity of T. asperellum isolated from cultures of mango (Tm), tomato (Tt) and onion (TC3) was evaluated against the pathogen through dual culture assays and medium enriched with culture filtrates of each T. asperellum isolate. The morphological characteristics and the molecular identification indicated that the SV1, SV2 and SV3 isolates correspond to Stemphylium vesicarium; whereas, the molecular analysis of the ITS sequences of the DC isolate shown similarity with fungi of the genus Alternaria. The infection with the isolates SV1, SV2 and SV3 of the leaves caused the formation of white spots, chlorosis, lesions of black color, the collapse, necrosis and death of the foliar tissue. The assay of antagonic activity with medium enriched with culture filtrates of each one of the isolates of T. asperellum indicated that the isolate TC3 of T. asperellum inhibited mycelial growth of S. vesicarium in 66.9%; followed by the isolates Tt and Tm with 43.5 and 37.2%, respectively. The TC3 and Tm isolates of T. asperellum also inhibited the production of conidia of S. vesicarium by 40 and 45%, respectively. The three isolates of T. asperellum inhibited the mycelial growth of S. vesicarium in dual culture; the Tm isolate shown the highest percentage of mycelial growth inhibition (61.1%), followed by the Tt and TC3 isolates of T. asperellum with 56.4 and 54.4%, respectively. In conclusion, on onion grown in the state of Puebla, Mexico one of the causal agents of leaf blight is S.
vesicarium and could be associated with Alternaria species. The TC3 isolate of T. asperellum obtained from onion cultures could be an alternative for the control of S. vesicarium. This is the first study that reports the presence of S. vesicarium on onion grown in Mexico and that for the identification of the species combines morphological and molecular data.
