La pérdida de vigor y la susceptibilidad al ataque de plagas de almacén de las semillas de maíz (Zea mays L.) con endospermo azul, constituyen serios inconvenientes para su producción masiva, dado que requiere de un manejo más escrupuloso. El deterioro de la calidad de las semillas se inicia en su madurez fisiológica, y ocurre mediante una serie de eventos físicos, fisiológicos, bioquímicos y moleculares, incluyéndose en éstos alteraciones en el metabolismo del ADN. En este estudio se examinaron los daños causados por el envejecimiento artificial en dos modalidades: calor seco (60 ºC durante 48 h) y calor húmedo (41 ºC, 100 % h. r., por 48 h), sobre las semillas de cuatro genotipos criollos de maíz azul (Oaxaca-711, Puebla-479, Cocotitlán y Cuijingo), producidos en el 2005. La calidad de las semillas fue evaluada empleando pruebas físicas (verde rápido), fisiológicas (germinación normal y protrusión radicular), bioquímicas (viabilidad, lixiviación de solutos y conductividad eléctrica) y además se obtuvo su perfil de bandeo genómico mediante la amplificación aleatoria del ADN polimórfico (RAPD, por sus siglas en inglés). Los resultados evidenciaron la respuesta genotípica particular de las variedades de maíz azul en cada uno de los análisis efectuados, lo cual se tradujo en un diverso grado de tolerancia al envejecimiento. Además, se concluyó que la RAPD es una metodología confiable para establecer la huella genómica de semillas de maíz. Cabe destacar que la caracterización varietal obtenida es robusta e integró diversas facetas del proceso germinativo, principalmente del análisis molecular, por lo que esta metodología pudiera ser una opción para que los programas de fitomejoramiento y de producción de semillas de maíz, identifiquen fehacientemente sus materiales genéticos.
ABSTRACT
The loss of vigor and the susceptibility to the attack of warehouse plagues of the maize seeds (Zea mays L.) with blue endosperm, they constitute serious inconvenience for its massive production, since it requires of a more scrupulous handling. The deterioration of the seeds quality begins in its physiological maturity, and occurs through a series of physical, physiological, biochemical and molecular events, including in these alterations in the metabolism of the DNA. In this study the damages caused by the artificial ageing in two modalities were examined: dry heat (60 ºC during 48 h) and humid heat (41 ºC, 100 % h. r., by 48 h), on the seeds of four native blue maize genotypes (Oaxaca-711, Puebla-479, Cocotitlán and Cuijingo), produced in 2005. The seeds quality was evaluated using physical tests (green fast), physiological (normal germination and root protrusión), biochemical (viability, leaching of solutes and electrical conductivity) and in addition was obtained its profile of banding genomic through the random amplified of the polymorphic DNA (RAPD). The results showed the particular genotypic response of the varieties of blue maize in each one of the conducted analyses, that which was translated in a diverse degree of tolerance to the ageing. In addition, it was concluded that the RAPD is a reliable methodology to establish the genomic fingerprint of maize seeds. It is necessary to highlight that the varietal characterization obtained is robust and integrated diverse facets of the germinative process, mainly of the molecular analysis, reason why this methodology could be an option for plant breeding programs and production of maize seeds, identify reliably their genetic materials.
