Comparación de las técnicas de gradiente de Percoll y Ficoll-Hipaque para el aislamiento de Linfocitos de Sangre Periférica Humana, Hospital Santa María del Socorro Ica

La investigación tiene como objetivo comparar las técnicas de gradiente de percoll y ficoll hipaque para el aislamiento de linfocitos de sangre periférica humana. Se diseñó un estudio prospectivo, transversal y analítico,dirigido a la evaluación de dos técnicas de concentración linfocitaria, para el cual se colectaron muestra de sangre de individuos sanos en tubos EDTA con sistema al vacío, basado en un muestreo probabilístico; posterior a la aplicación de ambas técnicas, se determinó el recuento de linfocitos, viabilidad celular e impurezas celulares de los concentrados, y se realizó análisis pareado para contrastar las hipótesis de estudio. Se evidenció un mayor recuento de linfocitos (p<0.001)obtenido por el método de ficoll-hipaque (10836±3269 células/μL) en comparación con el método de percoll (5608±3261 células/μL); en contraste a la viabilidad la cual fue mucho mayor (p<0.001) en el método por Percoll (71.3±15.3%), que el de Ficoll-Hipaque (31.0±23.8%) y finalmente el nivel de impureza celular obtenida por el método de percoll (36.9±17.9%), fue mayor (p<0.001) en comparación con el obtenido por el método de Ficoll-Hipaque (19.4±10.1%). Los resultados de recuento, viabilidad e impurezas celulares presentaron diferencias significativas entre las técnicas de ficoll-hipaque y percoll.
