Influência do Tamoxifeno associado a Cisplatina na osseointegraçao de implantes de titânio

O propósito do presente estudo foi avaliar a osseointegraçao de implantes de titânio instalados em tíbias de ratos modificados sistemicamente pelo Tamoxifeno (TAM) associado ou não a Cisplatina (CIS). Foram utilizadas 40 ratas, distribuídas em dois grupos experimentais (n=20), os animais receberam ovariectomia bilateral 8 semanas previas ao início do experimento, após 2 semanas do início receberam administração por gavagem de citrato de Tamoxifeno (TAM) diariamente uma vez por dia por todo o período experimental, após 6 semanas instalação dos implantes em tíbia bilateralmente. Dez animais de cada grupo foram eutanasiados na 7ª e 11ª semana, as tíbias direita e esquerda foram coletadas e análises histológicas e histométricas foram realizadas. O grupo solução salina (SS) apresentou maior contato osso implante (COI) e porcentagem de tecido ósseo neoformado (PTON) em todos os períodos experimentais quando comparado ao grupo CIS. A análise histológica dos tecidos periimplantares mostrou que o grupo TAM apresentou características histológicas que se mostraram similares ao grupo controle com maior quantidade de tecido ósseo periimplantar e menor quantidade de tecido conjuntivo. Apesar do efeito agonista do Tamoxifeno no osso, sua influência não é capaz de reverter os efeitos deletérios da Cisplatina. The purpose of the present study was to evaluate the osseointegration of titanium implants installed in the tibia of rats systemically modified by tamoxifen (TAM) associated or not with Cisplatin (CIS). Forty rats were used, distributed in two experimental groups (n=20), the animals received bilateral ovariectomy 8 weeks before the beginning of the experiment, after 2 weeks of the beginning they received administration by gavage of Tamoxifen Citrate (TAM) daily once a day for the entire experimental period, after 6 weeks of implant placement in the tibia bilaterally. Ten animals from each group were euthanized at the 7th and 11th week, the right and left tibias were collected and histological and histometric analyzes were performed. The saline solution (SS) group showed higher bone-implant contact (COI) and percentage os newly formed bone tissue (PTON) in all experimental periods when compared to the CIS group. The histological analysis of the peri-implant tissues showed that the TAM group presented histological characteristics that were similar to the control group, with a greater amount of peri-implant bone tissue and less connective tissue. Despite the agonist effect of Tamoxifen on bone, its influence is not capable of reversing the deleterious effects of Cisplatin.
