Eficiencia reductora de Pseudomonas sp. en concentraciones de cromo hexavalente

El objetivo fue evaluar la eficiencia reductora de Pseudomonas sp. en diferentes concentraciones de cromo hexavalente. La bacteria procedió del cepario del Laboratorio de Biotecnología e Ingeniería Genética de la Universidad Nacional de Trujillo, aislada de agua residual de curtiembre. El cultivo se reactivó en caldo nutritivo por 24 h a temperatura ambiente (19.5 ± 0.5ºC) y en agitación (150 rpm). Luego, se sembró en placas y se evaluó sus características bioquímicas. El cultivo stock se inoculó en medio mínimo de sales suplementado con glucosa al 10 % y pH 7. La incubación se realizó en matraces (150 mL) y las concentraciones de Cr (VI) fueron de 100 (control), 200, 400 y 800 ppm, cada uno por triplicado. En cada matraz se inoculó Pseudomonas sp. obtenida de la fase log (3 x 108 cél/mL) y se mantuvo en agitación (150 rpm) durante 120 h. El mayor crecimiento poblacional de Pseudomonas sp. a las 120 h de cultivo fue de 0.59 x 108 cél/mL y obtenido en el medio con 400 ppm de Cr (VI). La mayor eficiencia reductora de Pseudomonas sp. para el cromo hexavalente fue de 86.33 %, 85.08 % y 84.58 %, sin diferencias (p>0.05), y fue obtenido en 100, 200 y 400 ppm de Cr (VI), respectivamente The aim was to evaluate the reducing efficiency of Pseudomonas sp. in different concentrations of hexavalent chromium. The culture was reactivated in nutrient broth for 24 h at room temperature (19.5 ± 0.5°C) and under stirring (150 rpm). Then, it was plated and its biochemical characteristics were evaluated. The stock culture was inoculated in minimal medium of salts supplemented with 10 % glucose and pH 7. Incubation was carried out in flasks (150 mL) and Cr (VI) concentrations were 100 (control), 200, 400 and 800 ppm, each in triplicate. Pseudomonas sp. Was inoculated in each flask. obtained from the log phase (3 x 108 cell/mL) and kept under stirring (150 rpm) for 120 h. The highest population growth of Pseudomonas sp. at 120 h of culture it was 0.59 x 108 cell / mL and obtained in the medium with 400 ppm of Cr (VI).
