Perfil antigénico del fluido vesicular de Cysticercus tenuicollis determinado por Western Blot utilizando suero hiperinmune de Oryctolagus cuniculus

Abstract

Cysticercus tenuicollis es parásito del omento de ovinos y la larva cisticerco o metacestodo de Taenia hydatigena que reside en el intestino del perro. No se tiene información relacionada con el perfil inmunológico del fluido vesicular (FV) de este cisticerco, aspecto que podría ser de utilidad como fuente de antígenos alternativos para el diagnóstico de la cisticercosis humana, enfermedad causada por Cysticercus cellulosae y endémica en Sudamérica, tal como se ha hecho con los cisticercos de Taenia crassiceps y Taenia taeniformis. La presente investigación tuvo como objetivo determinar el número y el peso molecular las bandas inmunoreactivas del FV de C. tenuicollis mediante Western blot utilizando suero hiperinmune de Oryctolagus cuniculus “conejo” macho de raza Nueva Zelanda. Para ello, se recolectó ejemplares de C. tenuicollis del omento de ovejas parasitadas naturalmente y sacrificadas en el Camal de El Porvenir de Trujillo (Perú) y de ellos, el FV en asepsia. Los anticuerpos específicos se obtuvieron inmunizando a un conejo por vía intramuscular con el FV centrifugado, a una concentración de 1410 ug/mL, cinco veces semanalmente, la primera con adyuvante completo de Freud y las restantes con adyuvante incompleto. Se realizo la técnica de Western blot y se detectaron ocho bandas inmunoreactivas cuyos pesos moleculares, en kDa, fueron de: 76, 70, 44, 37, 25, 18, 11, 8. La banda de 18 kDa es la única coincidente con aquellas presentes en el FV de C. cellulosae y catalogadas como bandas inmunodiagnósticas.

Cysticercus tenuicollis is a parasite of the omentum of sheep and the metacestode or cysticercus of Taenia hydatigena that resides in the intestine of the dog. There is no information related to the immunological profile of the vesicular fluid (VF) of this cysticercus, an aspect that could be useful as a source of alternative antigens for the diagnosis of human cysticercosis, a disease caused by Cysticercus cellulosae and endemic in South America, such as has been done with the cysticerci of Taenia crassiceps and Taenia taeniformis. The objective of this research was to determine the number and molecular weight of the VF immunoreactive bands of C. tenuicollis by Western blot using hyperimmune serum from male Oryctolagus cuniculus "rabbit" of the New Zealand breed. For this, specimens of C. tenuicollis were collected from the omentum of sheep naturally parasitized and slaughtered in the slaughterhouse of El Porvenir in Trujillo (Peru) and from them, the VF in aseptic conditions. The specific antibodies were obtained by immunizing one rabbit intramuscularly with the centrifuged VF, at a concentration of 1410 ug/mL, five times weekly, the first with complete Freud's adjuvant and the rest with incomplete adjuvant. Using the Western Blot technique, eight immunoreactive bands were detected whose molecular weights, in kDa, were: 76, 70, 44, 37, 25, 18, 11, 8. The 18 kDa band is the only one that coincides with those present in the FV of C. cellulosae and cataloged as immunodiagnostic bands.
