Resumen
The purpose of this titration work was to determine the inhibitory activity of the alkaloidal fraction of Hymenocallis sp. on the enzyme’s acetylcholinesterase and butyrylcholinesterase, species belonging to the Amaryllidaceae family, through the process of maceration with methanol. The crude extract was obtained, alkaloidal extraction was performed based on protocols with changes in pH and use of organic solvents, thus obtaining a total alkaloid yield of 1.634 %. The alkaloids were identified by gas chromatography coupled to mass spectrometry. For the determination of the inhibitory activity on the enzyme’s acetylcholinesterase and butyrylcholinesterase, the colorimetric method of Ellman et al. (1961), modified by Ortiz et al. (2016) and galantamine was used as the reference compound, for the calculation of IC50. In the alkaloidal extract of Hymenocallis sp. four alkaloids were identified, the majority alkaloid was tazetine with a concentration of 66.3107 µg gal / 100 mg PS, followed by licorin with a 12.2823 µg gal / 100 mg PS. In a smaller amount, hemantamine is found with 10.3387 µg gal / 100 mg PS and lycoramine with the 10.0148 µg gal / 100 mg PS. In the inhibitory activity of the alkaloidal fraction of Hymenocallis sp. on the enzyme acetylcholinesterase and butyrylcholinesterase the IC50 was determined, using a variation in the concentrations of cosolvent A and cosolvent B. In this way the best IC50 was obtained in the stock solution 3 with an IC50 of 13.60 ± 1.55 µg/ml in AChE and BuChE the best IC50 of 166.30 ± 2.09 µg / ml was obtained in the stock solution l. The inhibitory activity of Hymenocallis sp. on the enzyme acetylcholinesterase is considered high, and on the enzyme butyrylcholinesterase, it is deemed to be medium. Further studies on Hymenocallis sp. due to its limited information and its alkaloidal content with pharmacological potential.
