Resumen
This study aims to isolate and identify the cultivable mycobiota associated with the phyllosphere of ten individuals of capulí (P. serotina Mc Vaugh) in the province of Chimborazo. Several morphotypes belonging to healthy and diseased leaves were obtained, as well as several methodologies were applied to reach this objective. Disinfection of leaves was carried out using tap water, alcohol (70%), sodium hypochlorite NaClO (1%) and sterile distilled water in the processing of samples, methodology of humid chamber and isolation of the tissue of a leaf was used for the isolation, later the morphotypes were preserved with the Castellani method. For the identification of morphotypes was carried out by microculture and printing technique, the Pantone color key was used for colorimetry and several graphics for the description of morphotypes in their edges, elevation, margins, texture, and pigmentation. For the analysis of the results in the case of the radial growth an average of the growth was carried out with 3 repetitions to obtain the average of each day, for the colorimetry and description of morphotypes was based on the above mentioned. Finally, for the diversity indexes, the program R Studio was used for the data analysis of the individuals. It is concluded that the total pure isolates were 87 (42 identified), from the healthy leaves were obtained 28 morphotypes non identified and 28 identified, on the other hand, the diseased leaves were obtained 17 morphotypes non identified and 14 identified, several of them belong to different genera. It is recommended to carry out analysis of the isolates, pathogenicity tests, investigations on the pigments produced and verification of endophytism by the fungi reported in healthy leaves.
