Carotenoids are pigments that are involved in photosynthesis, photoprotection and plant hormones synthesis. Carotenoid synthesis is induced by light through the signal transduction mechanism induced by photoreceptors in photosynthetic tissues, which leds to the expression of PSY, that codifies for phytoene synthase enzyme, the main point of regulation in carotenoid synthesis.
Daucus carota (carrot) accumulates high levels of carotenoids in its storage root that develops in darkness. Contrary to other plants, light inhibits storage root swelling, carotenoid synthesis and gene expression. To understand the molecular mechanisms behind this phenomenon, an RNA-seq analysis was carried out comparing roots grown in dark and in the presence of light. Interestingly, some light regulated genes, such as PHYTOCHROME INTERACTING FACTOR 3 (PIF3), were up-regulated in dark grown roots. In Arabidopsis, PIF3 codes for a transcription factor that represses photomorphogenesis and carotenoid synthesis in darkness. In carrot, DcPIF3 expresses mostly in dark grown roots during storage root development. In this thesis we determined that carrot DcPIF3 presents 48% of identity and 58% of similarity when compared to AtPIF3, and contains conserved DNA and protein binding domains. Its subcellular localization was found to be nuclear, according to the expected function. Arabidopsis DcPIF3 transgenic plants of 8 weeks, showed an elongated hypocotyl and a decrease in the levels of expression of AtPSY, together with a decrease in total carotenoids. Four day-old transgenic seedling that were subjected to grow four days in dark presented a longer hypocotyl, lower levels of AtPSY expression and total carotenoids. On the contrary, seedlings that were maintained 8 days under photoperiod, showed a reduced length in hypocotyl compared to wild type, and higher AtPSY transcript levels and carotenoid content. These results let us to conclude that DcPIF3 codifies for a functional factor that in darkness negatively regulates photomorphogenesis and carotenoid synthesis in Arabidopsis thaliana. En plantas, los carotenoides son pigmentos que participan en la fotosíntesis, la fotoprotección y la síntesis de hormonas vegetales. En tejidos fotosintéticos, la síntesis de carotenoides es inducida por la luz a través del mecanismo de transducción de señales inducido por los fotoreceptores induciendo un aumento en la expresión de PSY, gen que codifica para la enzima fitoeno sintasa y es el punto clave en la regulación de la síntesis de carotenoides.
Daucus carota (zanahoria) acumula altos niveles de carotenoides en su raíz de almacenamiento que se desarrolla en la oscuridad. Al contrario de otras plantas, la luz inhibe el desarrollo de la raíz, la síntesis de carotenoides y la expresión de genes carotenogénicos.
Para comprender los mecanismos moleculares detrás de este fenómeno, se realizó un RNA-seq comparando raíces cultivadas en la oscuridad y en presencia de luz. Sorprendentemente, algunos genes regulados por la luz, como el FACTOR DE INTERACCIÓN DE FITOCROMO 3 (PIF3), estaba regulado al alza en las raíces crecidas en oscuridad. En Arabidopsis, PIF3 codifica para un factor de transcripción que es activo en oscuridad y en esta condición reprime la fotomorfogénesis (induciendo el alargamiento del hipocotilo o tallo) y la síntesis de carotenoides.
En esta tesis de Magíster se caracterizó funcionalmente a DcPIF3. El análisis in silico mostró que DcPIF3 presenta un 48% de identidad y un 58% de similitud en comparación con AtPIF3, y que contiene dominios conservados de unión a ADN y proteínas. Se encontró que su localización subcelular era nuclear, de acuerdo con la función esperada. Plantas de Arabidopsis transgénicas de 8 semanas que expresan DcPIF3, mostraron un alargamiento del tallo, una disminución en la abundancia de transcritos de AtPSY y una disminución en los niveles de carotenoides totales.
Líneas transgénicas de DcPIF3 de 8 días sometidas a tratamiento de oscuridad por 4 días no mostraron diferencias en el largo del hipocotilo pero si menor nivel de expresión de AtPSY y menor concentración de carotenoides totales, al igual que clorofila a y b. Líneas transgénicas de DcPIF3 de 8 días crecidas bajo fotoperíodo mostraron un hipocotilo más largo, mayores niveles de expresión de AtPSY y una tendencia al alza en la concentración carotenoides totales, así no de clorofila a y b. Estos resultados permiten concluir que el gen DcPIF3 codificaría para un factor de interacción a fitocromo que en oscuridad estaría regulando negativamente la fotomorfogénesis y la síntesis de clorofilas y carotenoides en Arabidopsis thaliana.
