Resumen
Es sabido que la disminución de niveles de estrógenos durante la menopausia impacta negativamente en la remodelación ósea y la función vascular. Durante esta etapa, si bien los niveles circulantes de estradiol (E2) son considerablemente bajos, los de estrona (E1) se mantienen debido a su síntesis periférica, y representan la mayor fuente de estrógenos circulantes. E1 es el segundo estrógeno de producción ovárica con actividad biológica relevante. Si bien en trabajos previos hemos reportado que E1 exhibe una acción positiva sobre hueso promoviendo la osteoblastogénesis, también hayamos indicios de efectos negativos a nivel vascular tendientes a la promover la ateromatosis, cuya etapa más avanzada implica la calcificación vascular. Objetivo: El objetivo de este trabajo fue comparar los efectos osteogénicos de E1 sobre osteoblastos de calvaria y en células musculares lisas vasculares transdiferenciadas a linaje óseo; evaluando específicamente parámetros asociados a la diferenciación celular.Resultados: Como sistemas experimentales se emplearon cultivos primarios de osteoblastos (OB) de calvaria de ratas Wistar neonatas, aislados por digestión enzimática, y células musculares lisas vasculares obtenidas de aortas de ratas Wistar de 1 mes de edad inducidos a transdiferenciar a linaje óseo (CMLV-OB), por incubación durante 21 días en medio con β-glicerofosfato y calcio. Los OB y CMLV-OB fueron expuestos in vitro a E1 (10-8M) durante las últimas 48 h de cultivo. El estrógeno mostró un efecto mitogénico sobre ambos tipos celulares, estimulando la proliferación de las CMLV-OB y OB, evaluada por el ensayo de conversión de MTT (27% s/Cp<0,05 y 39% s/C p<0,05, respectivamente) y confirmada por conteo celular (28% s/C p<0,05 y 41% s/C p<0,05, respectivamente).La caracterización del modelo de transdiferenciacióndemostró que las CMLV-OB exhiben niveles elevados de actividad de FAL (medido en medio de cultivo y lisados de OB, usando ensayos enzimáticos) y depósito de calcio extracelular (tinción con rojo de Alizarina), similares a los de OB maduros. Al evaluar el efecto de E1, sobre las CMLV-OB observamos que el esteroide aumentó la actividad FAL (3,72±0,25 vs 3,00±0,14; E1 vs C, UI.10-2/mg proteína, p<0,001) y el número y tamaño de núcleos de calcificación en la matriz extracelular (56% s/C, p<0,05). En concordancia con este resultado, observamos descensos marcados del contenido de calcio en medio de cultivo (735,0±30,5 vs 468,2±21,1 C vs E1, µg calcio/mg proteína, método colorimétrico de o-cresolftaleincomplexona, p <0,001). Empleando la tinción de rojo Sirio estudiamos el efecto de la hormona sobre el depósito de colágeno extracelular. Se detectó un aumento significativo en el depósito de colágeno en CMLV-OB (↑19 % s/C, p<0,05). Resultados similares se obtuvieron en células nativas óseas (OB). El tratamiento con E1 estimuló la actividad FAL (4,64±0,32 vs 3,37±0,25 ; E1 vs C ; UI.10-2/mg proteína, p<0,001), el depósito de calcio (40,2% s/C, p<0,05) y de colágeno en la matriz extracelular (↑21 % s/C, p<0,05). En conclusión, los resultados presentados muestran una acción similar de E1en ambos sistemas celulares. Desde un punto de vista biológico los datos sugieren que la relevancia fisiológica es contrapuesta: una acción beneficiosa a nivel óseo favoreciendo la osteoblastogénesis y deletérea a nivel vascular promoviendo la calcificación vascular.
