Tephrocactus es un género de Cactaceae perteneciente a la subfamilia Opuntioideae, cuyas especies son, en su mayoría, endémicas de Argentina. Posee matas con ramas laxas, formadas por artejos globosos, frutos secos y semillas con arilo seminal blando arenquimático. La taxonomía del género ha sido inestable y controvertida. En este trabajo presentamos una hipótesis filogenética en base a datos moleculares (trnK-matK y psbA-trnH) que incluye el total de las especies de Tephrocactus, permitiendo evaluar la monofilia del género y discernir su posición dentro de Opuntioideae. Por otra parte, se obtuvieron los recuentos cromosómicos, cantidad de heterocromatina, genes de ADNr y contenido de ADN, de todas las especies. El número cromosómico en lasespecies del género es variable con 2n=22, 44, 77, 88, 242, 319. El contenido de ADN 2C mostró una correlación significativa y positiva con el nivel de ploidía y el número de genes de ADNr. En base al conjunto de nuevas evidencias moleculares, morfológicas y citogenéticas se proponen tendencias evolutivas de caracteres citogenéticos y morfológicos. Los caracteres más informativos para reconstruir la historia evolutiva del género son: número cromosómico, contenido de ADN, número de sitios ADNr, arbustos con articulaciones, hojas caducas, color de los tépalos, frutos secos y semillas con arilo seminal blando. La diversificación del género se asocia con tres eventos de poliploidía, lo que podría estar asociada a la reproducción vegetativa que permite en las especies mantener altos niveles de ploidia 2x=22 y 29.
