dc.descriptionMexico, with more than 25.000 different native plant germplasm, is known as center of germplasm diversity, and many of these are utilized as ornamental plants worldwide. Among different taxonomic families, some geophytic genera contrast due to the beauty of their flowers and the genetic variability found among their species including the genera Milla (Themidaceae), Sprekelia, Zephyranthes (Amarillydaceae) and Polianthes (Agavaceae). The identification of individual chromosomes is an useful tool in breeding programs, from the ploidy determination to introgression of chromosomal segments in interspecific hybrids. In the last decade, with the use of molecular cytogenetics, several species have been characterized; however, there are still few probes that can be used for this purpose in characterizing plant species. With the aim to identify individual chromosomes to assist in breeding programs of selected ornamental species, we have generated several probes from repetitive DNA by cloning and sequencing; these techniques are described. The usefulness of the probes is shown by the identification of single chromosomes through the use of molecular cytogenetics in different geophytes species with ornamental potential.
