A infecção pelo vírus Zika (ZIKV) tornou-se um grande problema de saúde pública após a descoberta em 2016 de que um aumento alarmante no número de recém-nascidos com microcefalia no Brasil poderia estar associado à ocorrência dessa doença viral durante a gravidez. A necessidade urgente de métodos simples de diagnóstico que permitissem um rastreamento rápido de casos suspeitos estimulou a busca de dispositivos de baixo custo capazes de detectar sequências específicas de ácidos nucléicos. O presente trabalho consistiu no desenvolvimento de filmes nanoestruturados formados por bicamadas de polímeros conjugados para a detecção rápida por fluorescência da presença do DNA do vírus Zika. Para isso, camadas alternadas de polianilina (PANI) e polipirrol (PPY) foram inicialmente crescidas sobre a superfície de folhas de polietileno tereftalato (PET). Os filmes obtidos foram então caracterizados por MEV, UV-vis, FTIR-ATR e medidas do ângulo de contato, e para sua utilização como supressores de fluorescência (“quenchers”) para o diagnóstico da Zika foi usada um cadeia simples de DNA, específica para o ZIKV e marcada com um fluoróforo (ssDNA- FAM). Para isso, a sonda ssDNA-FAM foi imobilizada na superfície das películas PANI-PPY/PET ou PPY-PANI/PET, do que resulta a extinção da fluorescência. Posteriormente, os filmes ao serem imersos em uma solução contendo o ssDNA alvo (fita de DNA complementar), ocorre a subsequente hibridização da sonda imobilizada no filme, com o seu alvo. Isso leva a uma alteração conformacional do DNA, que se desprende do filme, ocasionando a recuperação da fluorescência. Na busca da otimização das propriedades dos filmes nanoestruturados, investigamos tanto a cinética de imobilização quanto a de hibridização. Com isso, foi possível identificar o tempo necessário para que ocorra a saturação da interação entre o ssDNA-FAM e o filme (180 min) e também o tempo necessário para que aconteça a máxima hibridização entre o ssDNA-FAM e o DNA alvo (60 min). Com base nos parâmetros otimizados, estimamos os limites de detecção como sendo 345 pmol/L e 278 pmol/L para os filmes PPY- PANI/PET e PANI-PPY/PET, respectivamente. A simplicidade do procedimento, aliada ao fato de que uma resposta positiva/negativa pode ser obtida em menos de 60 minutos, sugerem que a proposta do uso desses filmes de bicamadas poliméricas é uma métodopromissor para o desenvolvimento de testes rápidos de diagnóstico molecular. CAPES Zika virus (ZIKV) infection became a major public health problem after the discovery in 2016 that an alarming increase in the number of newborns with microcephaly in Brazil could be associated with the occurrence of this viral disease during pregnancy. The urgent need for simple diagnostic methods that allow rapid screening of suspected cases has stimulated the search for low-cost devices capable of detecting specific sequences of nucleic acids. The present work consisted in the development of nanostructured films formed by bilayers of conjugated polymers for the rapid detection by fluorescence of the presence of Zika virus DNA. For this, alternating layers of polyaniline (PANI) and polypyrrole (PPY) were initially grown on the surface of polyethylene terephthalate (PET) sheets. The films obtained were then characterized by MEV, UV-Vis, FTIR-ATR and contact angle measurements, and for their use as quenchers for the diagnosis of Zika a single DNA strand specific for ZIKV, labeled with a fluorophore (FAM-ssDNA). For this, the FAM-ssDNA probe was immobilized on the surface of PANI-PPY/PET or PPY-PANI/PET films, resulting in a quenching of the fluorescence signal. Subsequently, the films were immersed in a solution containing the target ssDNA (complementary DNA strand), and the subsequent hybridization of the immobilized probe with its target leads to a conformational change of the DNA chain, which detaches from the film, causing fluorescence recovery. After investigating the kinetics of immobilization and the kinetics of hybridization, we optimized the properties of the nanostructured films. With this, it was possible to determine the time required for saturation of the interaction between FAM-ssDNA and the film (180min) and the time required for the maximal hybridization between FAM-ssDNA and target DNA (60 min). The corresponding detection limits were estimated as 345 pmol/L and 278 pmol/L for the PPY-PANI/PET and PANI-PPY/PET, respectively. The simplicity of the procedure coupled with the fact that a positive/negative response can be obtained in less than 60 minutes suggests that the proposal of using these polymeric bilayer films represents a promising methodology for the development of rapid molecular diagnostic tests.
