Diversidade de fungos conidiais em folhedo de mata ciliar em floresta úmida de Pernambuco

As matas ciliares produzem matéria orgânica que garantem nutrientes para a manutenção desse ecossistema, além de oferecer condições essenciais para o desenvolvimento dos fungos. Entre os fungos capazes de decompor a matéria orgânica estão os fungos conidiais, cuja principal importância para os ecossistemas é promover a ciclagem dos nutrientes a partir da decomposição. Diante disso, o presente trabalho teve como objetivo ampliar o conhecimento sobre a diversidade de fungos conidiais em folhedo de regiões de mata ciliar em áreas de Mata Atlântica sob proteção ambiental. Foram realizadas 12 expedições, entre agosto de 2014 e julho de 2016, em duas Unidades de Conservação (UC - REBIO Saltinho e RVS Gurjaú). Em cada UC foram selecionados seis locais para coleta do folhedo. As amostras de folhedo foi levada ao Laboratório de Hifomicetos de Folhedo da UFPE e submetidas à técnica de lavagem delicada em água corrente. O folhedo foi colocado em câmaras úmidas e incubado em temperatura ambiente. O material incubado foi observado em estereomicroscópio e microscópio de luz e examinados durante 30 dias. Outra parte do folhedo foi armazenada em freezer a -20 ºC para extração do ergosterol como estimador da biomassa de fungos. A inspeção diária de fragmentos foliares, ao microscópio, permitiu o registro de 177 táxons de fungos conidiais, sendo registrados 135 táxons na REBIO Saltinho com 1.385 ocorrências e 128 táxons no RVS Gurjaú com 1.276 ocorrências. Na REBIO Saltinho, os táxons predominantes foram Cryptophiale kakombensis, Beltrania rhombica, Beltraniella portoricensis, Helicomyces roseus, Subulispora procurvata e Wiesneriomyces laurinus; no RVS Gurjáu, os táxons predominantes foram C. kakombensis, B. rhombica, B. portoricensis e Perelegamyces parviechinulatus. Foram registradas três novas espécies, além de dois novos registros para a América do Sul. Os dados de constância foram semelhantes para a REBIO Saltinho e o RVS Gurjaú, nas duas áreas prevaleceram os táxons acidentais. A curva de acumulação de espécies produzida pelo estimador de riqueza Bootstrap permitiu a observação de aproximadamente 84% da riqueza estimada na REBIO Saltinho e 85% no RVS Gurjaú. A análise do folhedo ao longo de dois anos de coleta mostrou que não houve diferença de riqueza entre as áreas. A análise de agrupamentos mostrou resultados semelhantes entre as áreas, em que nenhuma estrutura foi encontrada para a comunidade de fungos, seja por pontos (espacial) ou períodos de coleta (sazonal). As duas primeiras componentes principais (PCA) extraídas explicaram 27,8% da variância dos dados. Foi observada uma forte relação positiva do teor de ergosterol contido no folhedo com a ocorrência de táxons na REBIO Saltinho 0,78, e uma relação forte negativa entre o teor de ergosterol com a ocorrência de táxons no RVS Gurjaú -0,76. O ergosterol é considerado um bom estimador para a biomassa dos fungos. Com as análises estatísticas, taxonômica e quantitativa foi possível observar que há uma alta diversidade de fungos conidiais nas áreas estudadas e que as mesmas apresentam uma forte semelhança, isso mostra a importância de estudar outras áreas do mesmo bioma para ampliar o conhecimento em regiões de floresta tropical úmida sob proteção ambiental. CAPES Riparian forests produce organic matter that guarantees nutrients for the maintenance of this ecosystem, besides offering essential conditions for the development of fungi. Among the fungi capable of decomposing organic matter are conidial fungi, whose main importance for ecosystems is to promote nutrient cycling from decomposition. Therefore, the present work had the objective of increasing the knowledge about the diversity of conidial fungi in leaf litter of riparian forest in areas of Atlantic Forest under environmental protection. Twelve expeditions were carried out, between August 2014 and July 2016, in two Conservation Units (UC - REBIO Saltinho and RVS Gurjaú). In each UC six sites were selected for leaf collection. The samples of leaf were taken to the Laboratory of Leaflitter Hyphomycetes of UFPE and submitted to the gentle washing technique under running water. The leaflet was placed in humid chambers and incubated at room temperature. The incubated material was observed under stereomicroscope and light microscope and examined for 30 days. Another part of the leaf was stored in a freezer at -20ºC for ergosterol extraction as a fungus biomass estimator. The daily inspection of leaf fragments under the microscope allowed 177 taxa of conidial fungi to be recorded, 135 taxa being recorded in REBIO Saltinho with 1,385 occurrences and 128 taxa in RVS Gurjaú with 1,276 occurrences. In the REBIO Saltinho, the predominant taxa were Cryptophiale kakombensis, Beltrania rhombica, Beltraniella portoricensis, Helicomyces roseus, Subulispora procurvata, Subulispora procurvata and Wiesneriomyces laurinus in RVS Gurjáu, the predominant taxa were C. kakombensis, B. rhombica, B. portoricensis and Perelegamyces parviechinulatus. Three new species were recorded, as well as two new registrations for South America. Constancy data were similar for REBIO Saltinho and RVS Gurjaú, in both areas accidental taxa prevailed. The species accumulation curve produced by the Bootstrap richness estimator allowed the observation of approximately 84% of the estimated richness in REBIO Saltinho and 85% in RVS Gurjaú. Leaf analysis over two years of collection showed that there was no richness difference between the areas. Grouping analysis showed similar results between areas where no structure was found for the fungal community, either by point (spatial) or collection periods (seasonal). The first two principal components (PCA) extracted explained 27.8% of the data variance. There was a strong positive relationship between the ergosterol content contained in the leaf and the occurrence of taxa in REBIO Saltinho 0.78, and a strong negative relationship between the ergosterol content and the occurrence of taxa in RVS Gurjaú -0.76. Ergosterol is considered a good estimator for fungal biomass. With the statistical, taxonomic and quantitative analyzes it was possible to observe that there is a high diversity of conidial fungi in the studied areas and that they have a strong similarity, this shows the importance of studying other areas of the same biome to expand knowledge in rainforest regions under environmental protection.
