A esquistossomose é considerada a segunda maior doença de importância á saúde pública dentre as doenças negligenciadas do mundo, onde o Praziquantel é o medicamento mais utilizado no mundo para o tratamento dessa doença, porém seu uso exclusivo pode ocasionar o desenvolvimento de uma possível resistência dos vermes de Schistosoma mansoni. Para aumentar o controle e a contenção da doença, novos fármacos vêm sendo desenvolvidos como alternativa para o tratamento, dentre eles o 2 - (- 5-bromo-1-h-indole-3-yl-methylene) -N- (naphthalene-1-ylhydrazine-carbothiamide (LQIT/LT-50) and 2- (1H-indole-3-methylene) - N - (naphthalen-1-yl) hydrazinocarbothioamide (LQIT/LT-51) demonstram excelente atividade esquistossomicida. Porém, essas moléculas apresentam baixa solubilidade em água, o que pode dificultar sua atividade farmacológica. Com isso, esse estudo teve como objetivo desenvolver dispersões sólidas à base de LQIT/LT-50 e LQIT/LT-51 para o tratamento da esquistossomose. Foram desenvolvidas dispersões sólidas utilizando Soluplus©, polietilenoglicol (PEG) e polivinilpirrolidona (PVP K-30) como carreadores hidrofilicos, obtidas pela técnica do solvente. As dispersões que obtiveram melhor resultado passaram por testes de solubilidade, espectroscopia de absorção na região do infravermelho com transformada de Fourier (FTIR), microscopia eletrônica de varredura (MEV), difratometria de raios-x (DRX), calorimetria diferencial exploratória (DSC), termogravimétria (TG), e espectroscopia Raman e atividade in vitro. As análises demonstraram que ao utilizar PVP-K30 obteve-se o melhor resultado, sugerindo que houve uma interação entre o PVP-K30 e os compostos analisados, observou-se assim incremento de solubilidade desses novas dispersões (LQIT/LT-50:PVP-K30 e LQIT/LT-51:PVP-K30), além da melhoria na atividade in vitro da dispersão de LQIT/LT-50:PVPK-30 1:4 (p/p), essa proporção mostrou resultados melhores quando comparado os resultados do LQIT/LT-50 isolado. Com isso podemos concluir que o desenvolvimento dessa dispersão se torna satisfatório para o desenvolvimento de novas formulações farmacêuticas, para o tratamento esquistossomicida. CAPES Schistosomiasis is considered the second major disease of public health importance among the neglected diseases in the world, where Praziquantel is the most used medicine in the world for the treatment of this disease, but its exclusive use may cause the development of a possible resistance of Schistosoma mansoni worms. To increase the control and containment of the disease, new drugs have been developed as an alternative to treatment, 2 - (- 5-bromo-1-h-indole-3-yl-methylene) -N- (naphthalene-1-ylhydrazine-carbothiamide (LQIT/LT-50) and 2- (1H-indole-3-methylene) - N - (naphthalen-1-yl) hydrazinocarbothioamide (LQIT/LT-51). demonstrate excellent schistosomicidal activity. However, these molecules have low water solubility, which can hinder their pharmacological activity. Thus, this study aimed to develop solid dispersions based on LQIT/LT-50 and LQIT/LT-51 for the treatment of schistosomiasis. Solid dispersions were developed using Soluplus©, polyethylene glycol (PEG) and polyvinylpyrrolidone (PVP K-30) as hydrophilic carriers, obtained by the solvent technique. The dispersions that obtained the best results were subjected to solubility tests, absorption spectroscopy in the infrared region with Fourier transform (FTIR), scanning electron microscopy (SEM), X-ray diffraction (XRD), differential scanning calorimetry (DSC), thermogravimetry (TG) and Raman spectroscopy and in vitro activity. The analyzes showed that when using PVP-K30 the best result was obtained, suggesting that there was an interaction between PVP-K30 and the analyzed compounds, an increase in solubility of these new dispersions was observed (LQIT/LT-50:PVP-K30 and LQIT/LT-51:PVP-K30), in addition to the improvement in in vitro activity of the LQIT/LT-50:PVPK-30 1:4 (w/w) dispersion, this proportion showed better results when comparing the results of the isolated LQIT/LT-50. With that we can conclude that the development of this dispersion becomes satisfactory for the development of new pharmaceutical formulations, for the schistosomicidal treatment.
