Objective: To evaluate the knowledge about sickle cell disease of health professionals through the construction and validation of the Knowledge in Sickle Cell Disease Instrument. Method: the study was carried out in five stages: 1) construction of the instrument: by querying the guidelines, protocols and manuals the identification of content about sickle cell disease and development of items of the instrument; 2) content validation by a Committee of judges: judges were asked to perform the assessment of each item of the instrument regarding the clarity and relevance of the content; 3) cultural adequacy (pre-test): this step was performed with a group of 36 health professionals working in the network of care; 4) analysis of reliability of the instrument by health professionals supported by the Intraclass correlation coefficient calculation. This step took place through the application of the instrument to a group of 300 professionals, of these, 153 accepted answer the instrument at the time test and, after 15 days, 102 attended the moment retest. 5) verification of knowledge of the 153 professionals participating in the analysis of process reliability. The data from this study were collected through the web and platform-Surv and tested on the statistical R programming environment. Results: the process of building the instrument resulted in the construction of 13 issues. In the evaluation of the judges, the instrument showed good acceptance, with content Validity index average of 0.88. During the pre-test found that 58% of workers reported some difficulty in answering the questions of number 11 and 13 because of the commands. This led to the change in the format of the commands that were placed in the spotlight in all items of the instrument. The analysis of the reliability of the instrument showed moderate agreement (0.67), indicating that the test-retest reproducibility is acceptable. In the analysis of the knowledge it was observed that the health professionals who have completed a course of distance education presented the score significantly higher DFConhecimento-45% [95% CI: 28% to 64%]-when compared with those who didn't arrive at the end of the course. Conclusion: the validation study contributed to provide a validated instrument from the point of view of content and reliable for the assessment of knowledge in sickle cell disease healthcare professionals the Brazilian context. In addition, the analysis with the results of DFConhecimento instrument has shown that the participation in the course of distance education about sickle cell disease have positive impact on the acquisition of knowledge about the disease on the part of health professionals.
