Evaluación de la calidad de ovocitos ovinos obtenidos mediante aspiración folicular empleando cuatro presiones de vacío

La técnica de aspiración folicular y obtención de ovocitos, consiste en extraer del ovario de una hembra por medio de aspiración las células germinales, logrando con ello poder obtener ovocitos viables para ser usados posteriormente para obtener embriones in vitro. No obstante, esta herramienta presenta algunas limitaciones en los procesos de producción in vitro de embriones, una parte fundamental es la presión que se ejerce en los procesos de aspiración folicular debido a que puede ser contraproducente en la calidad y en la morfología de los ovocitos.
El objetivo de este trabajo es evaluar la calidad de ovocitos ovinos a través de la implementación de cuatro presiones de vacío por medio de aspiración folicular y su efecto sobre la morfología ovocitaria. Para ello se utilizaron 5 ovarios por tratamiento de presiones de vacío, tratamiento 1= 25 milímetros de mercurio (mmHg), tratamiento 2= 35 milímetros de mercurio (mmHg), tratamiento 3= 45 milímetros de mercurio (mmHg), y tratamiento 4= 55 milímetros de mercurio (mmHg), evaluando la morfología, donde se determinó la cantidad de capas de células de la granulosa, si existieron variaciones en la forma o daño en la membrana celular.
Se realizó una prueba de análisis de varianza donde se estableció diferencias entre los tratamientos y las variables. Los tratamientos III y IV, presentaron el mayor número de ovocitos desnudos, sin embargo, los tratamientos I y II, presentaron la mejor morfología y calidad, estableciendo una diferencia significativa entre los tratamientos (p≤0.01) Las presiones de vacío en la aspiración folicular están relacionadas significativamente con la calidad de los ovocitos obtenidos. The technique of follicular aspiration and obtaining oocytes is part of the reproductive biotechnologies that can be used in the ovine species. It consists of obtaining from the ovary of a female by means of aspiration the germ cells of a genetically superior animal, thereby achieving to be able to obtain viable oocytes for later use to obtain embryos in vitro. However, this tool has some limitations in the processes of in vitro production of embryos, a fundamental part is the pressure exerted in the processes of follicular aspiration because it can be counterproductive in the quality and morphology of the oocytes.
With this in mind, the objective of this work is to evaluate the quality of sheep oocytes through the implementation of four vacuum pressures by means of follicular aspiration and their effect on ovocyte morphology.
For this, 5 ovaries per treatment will be used, which will be the vacuum pressures, treatment 1 = 25 millimeters of mercury (mmHg), treatment 2 = 35 millimeters of mercury (mmHg), treatment 3 = 45 millimeters of mercury (mmHg), and treatment 4 = 55 millimeters of mercury (mmHg), morphology evaluation , where the amount of granulosa cell layers is determined, if there are variations in the shape or damage in the cell membrane. A variance analysis test was performed where differences were found between the treatments and the variables. Treatments III and IV, location the largest number of naked oocytes, however, treatments I and II, the best morphology and quality, establishing a significant difference between treatments (p≤0.01) The vacuum pressures in follicular aspiration are related to the quality of the related oocytes.
