Implementation of a methodology to quantify microcirculatory parameters in cerebral tissue of a swine model in vivo

Maintenance of the microcirculatory function has been proposed as a key factor for the positive outcome of the performance of artificial oxygen carriers and plasma expanders. Such products are used to restore blood volume and oxygen carrying capacity in cases of hemorrhagic shock. The majority of the experimental data in which all this theory is based was obtained from a skin fold window chamber preparation that allows the direct visualization of the muscle and connective tissue microvessels. This vessels are thought to be the first tissues to be shot down during severe bleeding due to control mechanisms. For this reason the visualization and measurements should be done in the microcirculation of the main organs whose survival is highly compromised during hypoxia such as the brain, to contrast with the existing data. In this document a summary of a year of work for the implementation of a methodology for the direct visualization and quantification of microcirculatory parameters in cerebral tissue of a swine model is presented. La preservación de la función microvascular ha sido establecida como un factor clave en el desempeño final de cualquier transportador de oxigeno o expansor de plasma que pretenda ser usado para el restablecimiento del volumen sanguíneo y el transporte de oxígeno en casos de choque hemorrágico. La mayoría de los datos experimentales con los que se cuenta para soportar esta teoría han sido obtenidos mediante la visualización de la microvasculatura del tejido cutáneo, conectivo y muscular de pequeños animales implantados con una ventana cutánea dorsal, estos tejidos se conoce que son los primeros en ser apagados durante el progreso de un choque hemorrágico debido a los mecanismos de control homeostático, por esta razón se ha propuesto que la visualización y cuantificación de parámetros se debe hacer en los órganos principales como el cerebro. En este trabajo se presenta el resumen de un año de trabajo dedicado a la implementación de una metodología para la visualización directa y la cuantificación de parámetros microcirculatorios en el tejido cerebral de un modelo animal porcino.
