Orientadores: Maria Imaculada Zucchi, Pedro Henrique Santin Brancalion Tese (doutorado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: Atualmente, a restauração ecológica surge como uma atividade promissora e efetiva para recuperar a biodiversidade e os serviços ecossistêmicos onde eles foram perdidos e/ou reduzidos. A mata Atlântica é o bioma brasileiro onde os impactos oriundos dessas perturbações são mais acentuados. A diversidade genética torna-se importante para projetos de restauração, pois a sua variação pode afetar as taxas de endogamia e comprometer a perpetuação das espécies nativas reintroduzidas e, consequentemente, o sucesso da restauração, ao longo do tempo. Assim, os objetivos deste trabalho foram descrever o sistema reprodutivo de Myroxylon peruiferum L.f., avaliar a diversidade e estrutura genética da espécie em viveiros comerciais do estado de São Paulo e em populações naturais e restauradas na Mata Atlântica, com o auxílio de marcadores microssatélites e SNPs. Para o estudo do sistema reprodutivo foram coletadas sementes de uma área de remanescente natural de Mata Atlântica totalizando 200 sementes (20 sementes de cada árvore matriz). Para o estudo de diversidade genética em mudas de M. peruiferum, foram coletadas amostras em quatro viveiros comerciais. Para a comparação entre áreas naturais e restauradas, foram amostrados indivíduos em quatro populações contrastantes de Mata Atlântica (duas naturais e duas restauradas). Os resultados mostram que a população de M. peruiferum apresenta sistema reprodutivo misto e evidências de endogamia biparental. Faz-se necessária a coleta de sementes de pelo menos 47 árvores matrizes para fins de conservação e restauração florestal, para que se tenha uma amostra de sementes com tamanho efetivo que evite a depressão por endogamia. A diversidade genética das mudas de M. peruiferum foi baixa, indicando a necessidade de aperfeiçoar os sistemas de coleta de sementes e à compra de mudas de vários viveiros da mesma região para uso em um mesmo projeto de restauração. Os resultados das estimativas dos parâmetros populacionais sugerem que não existe diferença significativa em relação à diversidade genética e endogamia entre as populações naturais e restauradas indicando níveis similares de diversidade genética e que os projetos de restauração foram eficientes. Os resultados encontrados neste trabalho podem auxiliar pesquisadores e gestores em futuros projetos de conservação da espécie e uso na restauração ecológica Abstract: Currently, ecological restoration is emerging as a promising and effective activity to recover biodiversity and ecosystem services where they have been lost and/or reduced. The Atlantic Forest is the Brazilian biome where the impacts from these disturbances are more pronounced. Genetic diversity is important for studies of restoration, because its variation may affect the rates of inbreeding and compromise the perpetuation of the reintroduced native species and, therefore, the success of restoring in the long term. The objectives of this project were to describe the mating system of the Myroxylon peruiferum L.f.; to evaluate the genetic diversity and structuring of the species in commercial nurseries in the state of São Paulo and nature and restored populations in the Brazilian Atlantic Forest with microsatellite and SNPs markers. To study the mating system, seeds were collected from a natural remnants area of Brazilian Atlantic Forest, totaling 200 seedlings (20 seeds of each mother-tree). To study the genetic diversity in M. peruiferum seedlings, samples were collected in four commercial nurseries. To compare remnant and restored areas, individuals were sampled in four contrasting populations of the species (two natural areas and two restored areas). The results showed that the population of M. peruiferum presents a mixed-mating system and evidence the biparental inbreeding. We propose that at least 47 seed-trees are needed for seed collection efforts associated with conservation and forest restoration outcomes, which will provide a sample of seeds with an effective size that will enable the avoidance of an inbreeding depression. The genetic diversity of M. peruiferum seedlings was low, indicating the need to improve seed collection systems and buying seedlings of various nurseries in the same region for use in the same restoration project. The results of the population parameters estimates suggest that there is no significant difference of genetic diversity and endogamy among remnant and restored populations indicating similar levels of genetic diversity and the success of restoration projects.
