O uso da câmera Canon EOS 5S e a lente Infinity objetivo Probe TS 160 facilitaram a captura In situ das bandas Hunter-Schreger sem perda do foco Abstract: The Hunter-Schreger bands can be used to efficient biometric personal identification system, however the lack of technical development and standardization of protocols hinders the massive use of this tool. The aim of this work was to optimize the conditions for obtaining, processing and analyses of Hunter-Schreger bands images. The main drawbacks to obtain a good quality Hunter-Schreger bands image are: 1) the reflection of a percentage of the incident light caused by the curve and vitreous enamel surface,.2) the interruption of the prisms integrity by vertical cracks, 3) difficulty to capture the full pattern of Hunter-Schreger bands in a photograph 4) loss of focus by derivatives of physiological processes from individual movements. In order, a polarizing filter was used to reduce the reflection and the corn oil was used to reduce the loss of the light; 11 teeth were photographed using Canon EOS 5D camera adapted to a stereoscopic microscope using 1.3" and 2.0" exposure times. The contrast was measured with ImageJ software in two regions: (1) close to the area of intense shine and (2) after the crack. Data were analyzed using Wilcoxon in R software. Results showed that exposure time 1.3" in the region 1 and region 2 the polarized filter with the minima absorptions improved observation of Hunter-Schreger bands; the use of oil improved the visualization of Hunter-Schreger bands after the crack (p<0.05). The exposure time 2.0", showed no significant differences with polarizing filter (p>0.05), nevertheless greater contrast of the bands with the use of oil (p<0.05). To determine the optimal exposure time to corn oil, 5 teeth were photographed after 0, 1, 10 and 30 minutes of exposition. The contrast was measured with ImageJ software. ANOVA one way with repeated measures test (SPSS) showed that 1 minute exposure is sufficient for oil to penetrate the crack (p<0.05). To increase Hunter-Schreger bands viewing area were obtained two photographs of each tooth (test In situ: N=5, test In vivo: N=5) with lateral displacement of the incident light using Canon EOS 5S and objective probe TS Infinity 160. The images were contrasted in Corel PHOTO-PAINT or Imagemagick software. Areas of interest in the photographs were cut and added in Gimp. T-Test for paired samples (R) showed a highest number of minutiae (p<0.05) in the final images. The using Canon EOS 5S camera and objective Infinity Probe TS 160 facilitated the capture In situ of Hunter-Schreger bands without loss of focus Mestrado Histologia e Embriologia Mestra em Biologia Buco-Dental 33003033001P3 2014/07112-1 CAPES FAPESP
