Orientador: Alexandre Leite Rodrigues de Oliveira Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: Lesões axonais na interface do sistema nervoso central (SNC) e periférico (SNP), como a avulsão de raízes ventrais (ARV), induzem importantes processos degenerativos retrógrados, podendo resultar em morte neuronal e consequente perda da função motora. Nesse tipo de lesão ocorre a separação de raízes nervosas com a medula espinal, interrompendo o contato entre um motoneurônio e as fibras musculares por ele inervadas. Um tratamento promissor seria a utilização de selantes de fibrina, para reimplantar as raízes ventrais nos seus respectivos sítios de avulsão e assim, promover a reconexão do motoneuronio ao órgão alvo e consequentemente, o retorno funcional. Atualmente os selantes de fibrina são utilizados em uma vasta gama de procedimentos cirúrgicos, incluindo cirurgia cardiovascular, torácica, plástica e reconstrutiva, dentária e neurocirurgia. No presente estudo foram utilizados dois tipos de selantes de fibrina para reconectar as raízes ventrais no local exato da lesão. Um deles, produzido a partir do veneno de Crotalus durissus terrificus e outro produzido comercialmente, a fim de investigar a eficácia de ambos na regeneração nervosa e recuperação motora após avulsão de raízes ventrais medulares. Para isso, ratos Lewis fêmeas, com 10 semanas de idade, foram submetidos à avulsão de raízes ventrais medulares (L4-L6). Esses animais foram divididos em 3 grupos: 1) Avulsão de raízes motoras, sem reimplante; 2) Avulsão e reimplante das raízes motoras com selante de fibrina derivado do veneno de Crotalus durissus terrificus na superfície medular; 3) Avulsão e reimplante das raízes motoras com selante de fibrina comercial (Tissucol). A intumescência lombar e o nervo isquiático foram dissecados e os espécimes processados para a investigação da sobrevivência neuronal e contagem de axônios mielínicos, bem como análise de parâmetros histomorfométricos. A imunoistoquímica foi empregada para avaliar a estabilidade sináptica, reatividade astroglial e reatividade microglial, além da expressão de proteínas relacionadas ao processo regenerativo axonal. Adicionalmente, a evolução da recuperação motora, foi investigada através do método "walking track test" (CatWalk). Os resultados indicaram que o reimplante das raízes avulsionadas com ambos selantes de fibrina melhoraram significativamente a recuperação motora até 12 semanas pós-operatórias. Além disso, o número médio de axônios e a sobrevivência neuronal foram igualmente aumentados nos grupos onde houve o reimplante. Por fim, o reimplante promeveu preservacão parcial da cobertura sináptica e aumento da expressão de proteínas relacionadas ao processo de regeneracão axonal. Por outro lado, não houve diferenças na reatividade microglial e astrogliose reativa, na fase aguda pós-lesão, entre os grupos experimentais. Em conjunto, nossos resultados sugerem que o selante de fibrina derivado do veneno de Crotalus durissus terrificus, bem como o selante de fibrina comercial (Tissucol), utilizados para o reimplante de raízes medulares são similarmente eficientes, reimplantando as raízes avulsionadas de forma estável e duradoura, gerando efeitos neuroprotetores e promovendo melhora motora Abstract: Axonal injury in the interface of central nervous system (CNS) and peripheral (PNS) such as ventral roots avulsion (VRA) induce major retrograde degenerative processes which may cause neuronal death and loss of motor function. By this type of injury occurs the separation of nerve roots with the spinal cord, interrupting the contact between a motor neuron and the innervated muscle fibers. One promising treatment would be to use of fibrin sealants to redeploy the ventral root in their respective sites of avulsion, thereby promote the reconnection of motor neuron to the target organ, and therefore, return of functional. Currently fibrin sealants are used in a wide range of surgical procedures, including cardiovascular, thoracic, plastic and reconstructive, dental and neurosurgery. In the present research, we used two types of fibrin sealants to reconnect the ventral roots in the exact location of the injury. One of them, produced from the venom of Crotalus durissus terrificus other commercially produced to investigate the efficacy of both sealants to the nerve regeneration and motor recovery after avulsion of ventral spine roots. For this, Lewis rats female, 10 weeks old, were underwent to avulsion of ventral spinal root (L4-L6). These animals were divided in 3 groups: 1) avulsion of motor roots without reimplantation; 2) avulsion and reimplantation of motor roots with fibrin sealant derived from venom of Crotalus durissus terrificus on the spinal cord surface; 3) avulsion and reimplantation of motor roots with commercial fibrin sealant (Tissucol). The lumbar enlargement and sciatic nerve were dissected and samples processed for the investigation of neuronal survival and counting of myelinated axons as well as for analysis of histomorfometric parameters. Immunohistochemistry was used to evaluate synaptic stability, reactivity astroglial and microglial, beyond the expression of proteins related to axonal regeneration process. Additionally, the evolution of motor recovery was investigated using the "walking track test" (Catwalk). The results indicated that reimplantation of avulsion roots with both fibrin sealants significantly improved motor recovery up to 12 weeks postoperative. Moreover, the average number of axons and neuronal survival was also increased in the groups that received the reimplantation. Finally, reimplantation promoted partial preservation of synaptic coverage and increased expression of proteins related to axonal regeneration process. Furthermore, no differences in microglial reactivity and reactive astrogliosis in the acute phase after injury, among the experimental groups. Taken together, our results suggest that the derived fibrin sealant from venom of Crotalus durissus terrificus, as well as the commercial fibrin sealant (Tissucol), used for the reimplantation of spinal cord roots are similarly efficient, reimplanting the avulsion roots effectively, are neuroprotective and promoting the motor improvement Mestrado Biologia Celular Mestre em Biologia Celular e Estrutural 2013/04409-0 FAPESP
