Orientador: André Victor Lucci Freitas Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: O gênero Stalachtis pertence à família Riodinidae e apresenta borboletas aposemáticas com padrões alares miméticos. Essas borboletas ocorrem desde o Panamá até o sudeste do Brasil, sendo restritas à região Neotropical. As suas lagartas comem plantas da família Simaroubaceae com fitoquímicos tóxicos e de sabor amargo, sugerindo que as borboletas de Stalachtis possam ser impalatáveis. Nesse trabalho é proposta uma filogenia para Stalachtis com base em marcadores moleculares ¿ um gene mitocondrial (COI) e três genes nucleares: GAPDH, CAD e o RPS5. Foi investigada a distribuição geográfica das espécies do gênero Stalachtis e também foram delimitadas com base em evidências moleculares, biogeográficas e morfológicas. As análises de distância genética aqui utilizadas ¿ Neighbor-Joining, rede de haplótipos, distância genética ¿ mostraram padrões similares que levam as mesmas conclusões. As três análises filogenéticas feitas ¿ Máxima Parcimônia, Máxima Verossimilhança e Inferência Bayesiana ¿ recuperaram o gênero Stalachtis como monofilético (separado do grupo irmão Protonymphidia senta) e produziram topologias iguais no que se refere as relações internas do gênero. Os 100 indivíduos amostrados de Stalachtis se organizaram em 11 clados bem definidos e com alto valor de probabilidade posterior e de bootstrap, que podem ser atribuídos a 10 espécies diferentes. Todos os indivíduos se separam formando dois grandes clados irmãos. O primeiro clado inclui S. phegia + Stalacthis susanna stat. rev.. O segundo clado inclui todas as espécies restantes, com S. lineata aparecendo como grupo irmão das restantes. Estas outras se organizam em dois clados irmãos: 1) S. halloweeni + S. phaedusa (uma relação inesperada porém congruente com os padrões alares), e 2) S. euterpe, grupo irmão de S. magdalena, Stalacthis terpsichore stat. rev., S. calliope e uma nova espécie não descrita. A espécie revalidada Stalachtis susanna, apresenta várias características distinguíveis da espécie irmã S. phlegia, desde o padrão alar, caracteres na genitália masculina e números cromossômicos diferentes. Junto a isso, as análises moleculares propostas mostram essas espécies como entidades distintas (clados bem definidos nas filogenias, alta distância genética, rede de haplótipos desconectadas). Adicionalmente, temos as evidências biogeográficas, sendo S. phlegia uma espécie relacionada com a Floresta Amazônica e Cerrado, e S. susanna com a Mata Atlântica. Interessantemente, no clado de S. magdalena, os indivíduos se separaram em dois clados distintos e com alto suporte, mas não foi encontrada nenhuma evidência morfológica que sugira que existam duas espécies distintas. Essa divisão no DNA mitocondrial pode ter sido causada por: uma barreira no fluxo gênico; pela existência de duas espécies crípticas; ou pela seleção do genoma mitocondrial por parasitas citoplasmáticos. A segunda espécie aqui revalidada é S. terpsichore, que apresenta características morfológicas (genitália masculina e padrão alar) e dados genéticos que a definem como uma espécie válida. Além das espécies revalidadas, foram obtidos dados moleculares para a descrição de uma nova espécie de Stalachtis do Equador que apresenta diferenças genéticas e morfológicas de S. calliope. Não foi encontrada estruturação genética ou geográfica entre as subespécies amostradas, sugerindo que as subespécies atuais não são unidades evolutivas distintas Abstract: Stalachtis genus belongs to the Riodinidae family, these butterflies have aposematic wings with mimetic patterns. They can be found from Panama to the southeast of Brazil, being restricted to the Neotropics. Their caterpillars eat Simaroubaceae plants with toxic phytochemicals and bitter taste, suggesting that Stalachtis butterflies can be unpalatable. In this paper, we propose a phylogeny for Stalachtis based on molecular markers - one mitochondrial gene (COI) and three nuclear genes: GAPDH, CAD and RPS5. We investigated the geographic patterns of the species, and delimited this species based on molecular, morphological and biogeographic evidence. The genetic distance analysis used here - Neighbor-Joining, Minimum Spanning network, COI genetic distance - showed similar patterns that lead to the same conclusions. Three phylogenetic hypotheses were made - Maximum Parsimony, Maximum Likelihood and Bayesian Inference ¿ and they recovered the Stalachtis genus as monophyletic (separated from the sister group Protonymphidia senta) and produced the same topologies regarding the internal relationships between species. The 100 sampled individuals of Stalachtis organized into 11 clades with high posterior probability and bootstrapping, corresponding to 10 different species. All individuals are divided into two major sister clades. The first clade includes S. phegia + Stalacthis susanna stat. rev.. The second clade includes all other species, with S. lineata as a sister group to the other. These others are organized into two sister clades: 1) S. halloweeni + S. phaedusa (an unexpected relationship but congruent with the wing patterns), and 2) S. euterpe, sister group of S. magdalena, S. terpsichore stat. rev., S. calliope and a new undescribed species. The revalidated species Stalachtis susanna has several distinguishable characteristics from the sister species S. phlegia, such as the wing pattern, characters of male genitalia and different chromosome numbers. Furthermore, the molecular analysis proposals show these species as distinct entities (well-defined clades in the phylogeny, high genetic distance and disconnected haplotypes network). Additionally, we have the biogeographic evidence, because S. phlegia is more related to the Amazon and the Cerrado, and S. susanna with the Atlantic Forest. Interestingly, the S. magdalena individuals are divided into two distinct clades with high support, but we did not find morphological evidence to corroborate that there are two distinct species. This division in mitochondrial DNA may have been caused by a barrier in gene flow; the existence of two cryptic species; or by the selection of mitochondrial genome by cytoplasmic parasites. The second species revalidated S. terpsichore, presented morphological characters (male genitalia and wing patterns) and genetic data that define it as a valid species. In addition to the revalidated species, molecular data were collected to describe a new species of Stalachtis from Ecuador, which has genetic and morphological differences from the S. calliope.
