A classificação tribal de Rauvolfioideae tem sido tradicionalmente baseada em caracteres de frutos e sementes. Tabernaemontaneae é a maior tribo de Rauvolfioideae e foi recentemente dividida em duas subtribos, Ambelaniinae e Tabernaemontaninae. Trabalhos de caracterização de frutos e sementes, em conjunto com estudos florais, são fundamentais para auxiliar na compreensão da evolução de caracteres em Apocynaceae. O estudo objetivou caracterizar as sementes de onze espécies de Tabernaemontaneae, quatro de Ambelaniinae e sete de Tabernaemontaninae, visando a identificar caracteres estruturais taxonomicamente úteis, em sementes maduras, e a origem da excrescência carnosa, das dobras do tegumento e da ruminação do endosperma, em sementes jovens. As sementes foram coletadas e processadas conforme técnicas usuais de microscopia de luz e eletrônica de varredura. Os caracteres estruturais diagnósticos de Ambelaniinae são sementes nuas com exotesta formada por células em paliçada, mesotesta parcialmente comprimida, hilo elíptico/circular subterminal ventral, endosperma não ruminado e embrião linear. As espécies de Tabernaemontaninae possuem sementes recobertas por excrescência carnosa, tegumento reduzido à exotesta sinuosa e homogênea, com células que variam na dimensão radial, hilo alongado e localizado em um sulco longitudinal na face rafeal, endosperma ruminado e embrião espatulado. A testa sinuosa está presente em sementes de Ambelaniinae e de Tabernaemontaninae e se origina de forma semelhante, respectivamente, nas sementes dos gêneros Tabernaemontana e Spongiosperma. O desenvolvimento do endosperma é tardio em ambos os gêneros e torna-se ruminado apenas em Tabernaemontana. A excrescência carnosa que envolve as sementes de Tabernaemontana tem origem placentária e não funicular, como registrado na literatura. O estrato gelatinoso e translúcido que recobre a semente de Macoubea guianensis é uma parede celular com mucilagem e não um arilo. Essa evidência auxilia no entendimento da evolução dos caracteres em Tabernaemontaneae, pois sugere o aparecimento do arilo apenas no ancestral comum mais recente (ACMR) de Tabernaemontaninae. Nossos dados também sugerem que sementes com mucilagem podem ter dado origem a sementes nuas e estas, por sua vez, terem originado sementes com excrescência carnosa de origem placentária no ACMR do clado Tabernaemontaneae s. str. (clado Tabernaemontaninae Abstract: Apocynaceae is one of the largest families of angiosperms, with a remarkable variation in morphological traits. The tribal classification of one of its subfamilies, Rauvolfioideae, has been traditionally based on fruit and seed characters. Tabernaemontaneae is the largest tribe of Rauvolfioideae and is currently divided in two subtribes, Ambelaniinae and Tabernaemontaninae. Studies focused on the characterization of fruits and seeds, together with floral studies, are critical to help in understanding the evolution of characters in Apocynaceae. This study aims to characterize the seed morphology and anatomy of eleven species of Tabernaemontaneae, four from subtribe Ambelaniinae and seven from subtribe Tabernaemontaninae, in order to detect characters with taxonomic significance in mature seeds and to study the origin of fleshy excrescence, the folds of the seed coat, and endosperm rumination in young seeds. Seeds were collected, prepared and analyzed with usual techniques in light microscopy and SEM. All species of Ambelaniinae have naked seeds with a circular or elliptic hilum, exotesta formed by the palisade cells, mesotesta partially crushed, non-ruminate endosperm, and linear embryo. In species of Tabernaemontaninae, seeds are covered by placental outgrowths, with seed coat reduced to the tortuous and homogeneous exotesta having cells ranging in radial length, an elongated hilum in a longitudinal groove in the raphe side, ruminate endosperm and spatulate embryo. The infoldings and furrows of the seed surface develop in the same way in Spongiosperma spp. (Ambelaniinae) and Tabernaemontana spp. (Tabernaemontaninae). The endosperm develops late in both, but does not acquire a ruminate configuration in Tabernaemontana. In Tabernaemontana, the pulpy structure has placental origin, not funicular, as registered in the literature. Our results show that the viscous, translucent stratum covering the seeds of Macoubea guianensis is not an aril, but a cell wall with mucilage. This interpretation helps understanding the character evolution in the tribe by the suggestion that the aril arose only once in the most recent common ancestor (MRCA) of the Tabernaemontaninae. Our data also suggest that mucilaginous seed may give raise to naked seeds and these ones to the fleshy placental outgrowth in the MRCA Tabernaemontaneae s. str. clade (Tabernaemontaninae clade) Mestrado Biologia Vegetal Mestra em Biologia Vegetal 131920/2013-8 CNPQ
