Resumen
The effects of propranolol and atenolol were investigated on arterial hypertension, cardiomyocyte hypertrophy, and ventricular ischaemic lesions induced by an 8-week treatment with the nitric oxide synthase inhibitor Nω-nitro-L-arginine methyl ester (L-NAME; 20 mg/rat per day) in Wistar rats. Propranolol and atenolol (30 mg/rat per day each) were given in the drinking water concomitantly to L-NAME. Treatment with L-NAME induced marked arterial hypertension and cardiomyocyte hypertrophy, both of which were significantly reduced by propranolol and atenolol. A marked repairing fibrosis was also observed in L-NAME-treated rats and this was significantly attenuated in animals receiving the β-blockers. In L-NAME group, 33% mortality was observed, whereas all the animals from the other groups survived. Our study demonstrates that propranolol and atenolol reduce arterial hypertension, cardiomyocyte hypertrophy and myocardial fibrosis induced by L-NAME, suggesting that β-blockers are of beneficial value in treatment of vascular and cardiac alterations caused by chronic nitric oxide deficiency. 39 2 201 207 Moncada, S., Palmer, R.M., Higgs, E.A., Nitric oxide: Physiology, pathophysiology, and pharmacology (1991) Pharmacol Res, 43, pp. 109-142 Zatz, R., Baylis, C., Chronic nitric oxide inhibition model six years on (1998) Hypertension, 32, pp. 958-964 Ribeiro, M.O., Antunes, E., De Nucci, G., Chronic inhibition of nitric oxide synthesis: A new model of arterial hypertension (1992) Hypertension, 20, pp. 298-303 Erley, C.M., Rebmann, S., Strobel, U., Effects of antihypertensive therapy on blood pressure and renal function in rats with hypertension due to chronic blockade of nitric oxide synthesis (1995) Exp Nephrol, 3, pp. 293-299 Navarro-Cid, J., Maeso, R., Rodrigo, E., Renal and vascular consequences of the chronic oxide synthase inhibition: Effects of antihypertensive drugs (1996) Am J Hypertens, 9, pp. 1077-1083 K-Laflamme, A., Foucart, S., Moreau, P., Sympathetic functions in NG-nitro-L-arginine methyl ester-induced hypertension: Modulation by the renin-angiotensin system (1998) J Hypertens, 16, pp. 63-76 Moreno Jr., H., Nathan, L.P., Costa, S.K.P., Enalapril does not prevent the myocardial ischemia caused by the chronic inhibition of nitric oxide synthesis (1995) Eur J Pharmacol, 287, pp. 93-96 Akuzawa, N., Nakamura, T., Kurashina, T., Antihypertensive agents prevent nephrosclerosis and left ventricular hypertrophy induced in rats by prolonged inhibition of nitric oxide synthesis (1998) Am J Hypertens, 11, pp. 697-707 Luvara, G., Pueyo, M.E., Phillippe, M., Chronic blockade of NO synthase activity induces a proinflammatory phenotype in the arterial wall: Prevention by angiotension II antagonism (1998) Arterioscler Thromb Vasc Biol, 18, pp. 1408-1416 Verhagen, A.M., Hohbach, J., Joles, J.A., Unchanged cardiac angiotensin II levels accompany losartan-sensitive cardiac injury due to nitric oxide synthase inhibition (2000) Eur J Pharmacol, 400, pp. 239-247 Oliveira, C.F., Nathan, L.P., Metze, K., Effect of Ca2+ channel blockers on arterial hypertension and heart ischemic lesions induced by chronic inhibition of nitric oxide synthesis in the rat (1999) Eur J Pharmacol, 373, pp. 195-200 Van den Meiracker, A.H., Veld, A.J., Boomsma, F., Hemodynamic and β-adrenoceptor adaptations during long-term β-adrenoceptor blockade: Studies with acebutolol, atenolol, pindolol and propranolol in hypertensive patients (1989) Circulation, 80, pp. 903-914 Kahönen, M., Arvola, P., Mäkynen, H., Antihypertensive therapy and arterial function in experimental hypertension (1996) Gen Pharmacol, 27, pp. 221-228 Weibel, E.R., Stereological methods.
