Resumen
Introduction: The study analyzed positivity of polymerase chain reaction (PCR) on detection of DNA from Leishmania in patients' samples. Methods: Extracted DNA was submitted to L150/L152, 13Y/13Z, and seminested PCR (snPCR). Results: Results were evidenced by bands of approximately 120, 720, and 670 bp for L150/L152, 13Y/13Z, and snPCR, respectively. L150/L152, 13Y/13Z, and snPCR positivity indexes were 76.9, 56.4, and 69.2 (p>0.05), respectively, for suspected and 93.7, 68.7, and 84.4 (p<0.05), respectively, for confirmed. Conclusions: Preliminary results showed that these assays, mainly L150/L152 and snPCR, can detect Leishmania DNA and carry potential on laboratory diagnosis of leishmaniasis. 45 2 257 259 Fagundes, A., Schubach, A., Paula, C.C., Bogio, A., Antonio, L.F., Schiavoni, P.B., Evaluation of polymerase chain reaction in the routine diagnosis for tegumentary leishmaniasis in a referral centre (2010) Mem Inst Oswaldo Cruz, 105, pp. 109-112 Weigle, K.A., Labrada, L.A., Lozano, C., Santrich, C., Barker, D.C., PCR-Based diagnosis of acute and chronic cutaneous leishmaniasis caused by Leishmania (Viannia) (2002) J Clin Microbiol, 40, pp. 601-606 Parvizi, P., Mauricio, I., Aransay, A.M., Miles, M.A., Ready, P.D., First detection of Leishmania major in peridomestic Phlebotomus papatasi from Isfahan province, Iran: Comparison of nested PCR of nuclear ITS ribosomal DNA and seminested PCR of minicircle kinetoplast DNA (2005) Acta Tropica, 93, pp. 75-83 Paiva, B.R., Secundino, N.F.C., Nascimento, J.C., Pimenta, P.F.P., Galati, E.A.B., Andrade Jr., H.F., Detection and identification of Leishmania species in Field-captured phlebotomine sandflies based on mini-exon gene PCR (2006) Acta Tropica, 99, pp. 252-259 Pilatti, M.M., Ferreira, A.S., Melo, M.N., Andrade, A.S.R., Comparison of PCR methods for diagnosis of canine visceral leishmaniasis in conjunctival swab samples (2009) Res Vet Sci, 87, pp. 255-257 Aransay, A.M., Scoulica, E., Tselentis, Y., Detection and identification of Leishmania DNA in naturally infected sand flies by Seminested PCR on minicircle kinetoplastic DNA (2000) Appl Environ Microbiol, 66, pp. 1933-1938 Noyes, H., Reyburn, H., Bailey, J.W., Smith, D., A nested-PCR-based schizodeme method for identifying Leishmania kinetoplast minicircle classes directly from clinical samples and its application to the study of the epidemiology of Leishmania tropica in Pakistan (1998) J Clin Microbiol, 36, pp. 2877-2881 Rodgers, M.R., Popper, S.J., Wirth, D.F., Amplification of kinetoplast DNA as a tool in the detection and diagnosis of Leishmania (1990) Exp Parasitol, 71, pp. 267-275 Marques, M.J., Volpini, A.C., Coelho, G.L.L.M., Pinto, J.M., Costa, C.A., Mayrink, W., Comparison of PCR with other laboratorial methods for the diagnosis of American Cutaneous Leishmaniasis (2006) Diagnostic Microbiol Infect Dis, 54, pp. 37-43 Volpini, A.C., Marques, M.J., dos Santos Lopes, S., Machado-Coelho, G.L., Mayrink, W., Romanha, A.J., Leishmania identification by PCR of Giemsa-stained lesion imprint slides stored for up to 36 years (2006) Clin Microbiol Infect Dis, 12, pp. 793-821 Cheng, S., Fockler, C., Barnes, W.M., Higuchi, R., Effective amplification of long targets from cloned inserts and human genomic DNA (1994) Proceed Nat Acad Sci USA, 91, pp. 5695-5699 Rocha, L.S., Santos, C.B., Falqueto, A., Grimaldi Jr., G., Cupolillo, E., Molecular biological identification of monoxenous trypanosomatids and Leishmania from antropophilic sand flies (Diptera Psychodidae) (2010) Southeast Braz Parasitol Res, 107, pp. 465-468. , in Saini, R., Santhanam, J., Othman, N.H., Saini, D., Tang, T.H., Single-tube Seminested PCR Assay for detecting Human Papillomavirus in Clinical Samples (2009) Open Microbiol J, 3, pp. 106-112
